The CD ssDNA Index Kit for Illumina was created specifically for the Illumina CD ssDNA-seq Lib Prep Kit and CD Methyl-DNA Lib Prep Kit. The kit includes two kits for preparing DNA libraries for Illumina platforms. A Universal PCR Primer and 48 different 8bp indexed PCR primers are included in each kit. Different specimens can be distinguished using single-indexed primers. The kit is ideal for creating NGS libraries from denatured DNA, viral DNA, ancient DNA that has been highly degraded, and other single-stranded DNA samples. The ssDNA kit's workflow is straightforward: make libraries in two steps, then PCR and cleanup.

16-Hour Incubation: A 50 ul reaction system containing 5 ul of Oligos and 1 μg of Hind III-λ DNA incubated at 37 °C for 16 hours resulted in no band degraded detected by agarose gel electrophoresis. A 50 μl reaction system containing 5 ul of Oligos and 1 μg of T3 DNA incubated at 37 °C for 16 hours resulted in no band degraded detected by agarose gel electrophoresis.
Endonuclease Activity: A 50 ul reaction system containing 5 ul of DNA adapters and 1 ug of pX174RF I DNA incubated at 37°C for 4 hours resulted in < 10% conversion to RF Il analyzed by agarose gel electrophoresis.
