Real-time polymerase chain reaction (real-time PCR), also known as quantitative polymerase chain reaction (qPCR), is a sensitive, powerful, and low-cost method used to amplify and simultaneously monitor, and quantify or semi-quantify targeted DNA molecules, which is a conventional technology in biology labs to provide insights into gene expression and phenotypes of microorganisms on the genetic level. Studies of the microbial communities by qPCR methods have revealed striking similarities as well as differences in the ratio of certain species in specific groups of people that of different ages, diet habits, and health conditions. There are two detection methods of qPCR, the first is based on sequence-specific probe served as fluorescent signal report molecules; the second is based on generic non-sequence-specific double-stranded DNA-binding dye such as SYBR green.
