Double digest RAD sequencing (ddRAD-seq) is the RAD-derived sequencing strategy in which genomic DNA is digested by combining a restriction endonuclease with a common enzyme that recognises fewer sites and another enzyme that recognises more sites, eliminating the need for physical interruption and allowing direct screening of the target fragment and the introduction of an individual recognition tag (index) near the second site by PCR amplification, allowing more samples to be mixed together for high-throughput sequencing, a research method widely used for SNP discovery and genotyping.
